    Regulation of Organelle Transport    
    In Drosophila Cell Culture    
             
    One of the advantages of using Drosophila cells is the effectiveness of RNA interference (RNAi). Of all the proteins that we tested so far, the target proteins were consistently reduced to less than 10% of the control level without affecting other proteins. By combining the RNAi technology and high-resolution microscopy, individual components that are involved in transport and transport regulation can be identified. Furthermore, a novel preparation of Drosophila S2 cells was developed to generate polarized microtubule arrays in living cells without the interference of actin filaments, and thus allows us to dissect the microtubule-based transport machinery directly.
             
    S2 cell lines with organelles (peroxisome, lysosome, mitochondria) labeled with different fluorescent proteins have been generated in the lab. We are using these cell lines to study mechanisms of intracellular transport and its regulations.
